Oleuropein Promotes Neural Plasticity and Neuroprotection via PPARα-Dependent and Independent Pathways

چکیده
Oleuropein (OLE), a main constituent of olives, displays pleiotropic beneficial dynamic in health and disease; the effects are based mainly on its antioxidant hypolipidemic properties, capacity to protect myocardium during ischemia. Furthermore, OLE activates peroxisome proliferator-activated receptor (PPARα) neurons astrocytes, providing neuroprotection against noxious biological reactions that induced following cerebral The current study investigated effect regulation various neural plasticity indices, emphasizing role PPARα. For this purpose, 129/Sv wild-type (WT) Pparα-null mice were treated with for three weeks. findings revealed chronic treatment up-regulated brain-derived neurotrophic factor (BDNF) TrkB prefrontal cortex (PFC) via activation ERK1/2, AKT PKA/CREB signaling pathways. No similar observed hippocampus. OLE-induced BDNF appear be mediated by PPARα, because no alterations PFC mice. Notably, did not affect factors NT3 NT4/5 both brain tissues. However, fenofibrate, selective PPARα agonist, mice, whereas drug sites tested. Interestingly, provided differentiated human SH-SY5Y cells β-amyloid H2O2 toxicity independently from activation. In conclusion, drugs, acting either as agonists or independent mechanisms, could improve synaptic function/plasticity lesser extent hippocampus, thus beneficially affecting cognitive functions.

Farnesol, an isoprenoid, improves metabolic abnormalities in mice via both PPARα-dependent and -independent pathways.
Peroxisome proliferator-activated receptors (PPARs) control energy homeostasis. In this study, we showed that farnesol, a naturally occurring ligand of PPARs, could ameliorate metabolic diseases. Obese KK-Ay mice fed a high-fat diet (HFD) containing 0.5% farnesol showed significantly decreased serum glucose level, glucosuria incidence, and hepatic triglyceride contents. Farnesol-containing HFD ...
متن کاملLoss of sclerostin promotes osteoarthritis in mice via β-catenin-dependent and -independent Wnt pathways
INTRODUCTION Sclerostin is a Wnt inhibitor produced by osteocytes that regulates bone formation. Because bone tissue contributes to the development of osteoarthritis (OA), we investigated the role of sclerostin in bone and cartilage in a joint instability model in mice. METHODS Ten-week-old SOST-knockout (SOST-KO) and wild-type (WT) mice underwent destabilization of the medial meniscus (DMM)....
متن کاملREST suppression mediates neural conversion of adult human fibroblasts via microRNA‐dependent and ‐independent pathways
Direct conversion of human fibroblasts into mature and functional neurons, termed induced neurons (iNs), was achieved for the first time 6 years ago. This technology offers a promising shortcut for obtaining patient- and disease-specific neurons for disease modeling, drug screening, and other biomedical applications. However, fibroblasts from adult donors do not reprogram as easily as fetal don...
متن کاملSecretoneurin promotes neuroprotection and neuronal plasticity via the Jak2/Stat3 pathway in murine models of stroke.
Secretoneurin (SN), a neuropeptide derived from secretogranin II, promotes neurite outgrowth of immature cerebellar granule cells. SN also aids in the growth and repair of neuronal tissue, although the precise mechanisms underlying the promotion of brain tissue neuroprotection and plasticity by SN are not understood. Here, in a rat model of stroke and in ischemic human brain tissue, SN was mark...
متن کاملPharmacological evidence for lithium-induced neuroprotection against methamphetamine-induced neurodegeneration via Akt-1/GSK3 and CREB-BDNF signaling pathways
Objective(s): Neurodegeneration is an outcome of Methamphetamine (METH) abuse. Studies have emphasized on the neuroprotective properties of lithium. The current study is designed towards evaluating the role of Akt-1/GSK3 and CREB-BDNF signaling pathways in mediating lithium neuroprotection against METH-induced neurodegeneration in rats. Materials and ...
